Chasing a target of 259 runs, the hosts won the game with more than two overs to spare.
England scored 258-8 in their innings, with Joe Denly top-scoring with 87.
In response, South Africa captain Quinton de Kock hit 107 to help his team register a comfortable victory.
The second match takes place on Friday in Durban.
